You may ask how this is possible, how can these ingredients deliver such wide-reaching effects? The secret sauce is in their mechanism of action. Many people may be aware of an ingredient which has been around the supplement industry for as far back as we can remember, but never really delivered much at all. The Science Behind of ALPHABULK:

OL introduces a completely new approach to boosting cAMP in skeletal muscle through targeting a novel pathway known as the ORs. They are predominantly expressed by the main olfactory epithelium at nasal level and are responsible for the generation of smell, however, according to developing research over recent years, it has been found that they are also found in many different areas of the body including skeletal muscle, the liver, heart, brain and fat cells and performing some very interesting and important functions.

Before we dive into the individual ingredients in the formula, let’s talk about cAMP, a very important secondary messenger for signal transduction at the cellular level which has been shown to perform key functions in the regulation of skeletal muscle mass and function. This occurs through enhancing the expression of specific target genes, which in turn regulates the functions of a series of protein targets including ion channels, enzymes, and transcription factors. In fact, cAMP-elevating agents have been reported to promote significant increases in total protein synthesis as well as reductions catabolic processes in skeletal muscles.

Increasing levels of cAMP can also support increased levels of lipolysis and oxidation of fatty acids, it is also a key molecule in the regulation of glycogen synthesis and importantly the synthesis of key anabolic hormones such as testosterone through enhancement of Leydig cell function.

α-Cedrene exerts its anabolic effects due to its ability to act as an agonist to the Human OR subtype known as OR10J5, which leads to the activation of a downstream signalling pathway cAMP-protein kinase A-cyclic AMP-responsive element-binding protein (CREB) in the skeletal muscle. This same pathway applies to other areas of the body such as in the liver and in fat cells whereby improved glucose tolerance, enhanced insulin sensitivity and enhanced fat lipolysis and oxidation have been noted in research.

Interestingly, α-Ionone appears to exert its potent effects through a different OR subtype known as OR51E2. The downstream effects appear similar to that of OR10J5 and we can expect to effects detailed above, given it is through a slightly different mechanism of action, we suspect that the combination of the two ingredients will have a 1+1 = 3 effect, leading to far greater elevations in cAMP compared to either individually. We fully intend to achieve maximal effects given we have opted for full clinical dosages of both ingredients! 

It is clear that both α-Cedrene and α-Ionone are extremely potent agents in their own right, but why stop there? we wanted to really drive this formula to new heights and deliver unquestionably powerful effects so we opted to include additional ingredients which will potently synergise with elevations in cAMP to amplify the overall effects of the product.

What better choice then Tectochrysin, which is incredibly potent anabolic ingredient in its natural form, but Olympus Labs never settle for good, we strive for outstanding. Olympus Labs utilized its custom PhytoFUSE delivery system, to help improve the bioavailability of Tectochrysin dramatically! Not only does it significantly aid in the oral absorption, it also increases the serum half-life of the compound, allowing a longer duration of action. With the above in mind, let's dive into more detail on each of the ingredients, outlining how they play their role in the formula:

α-Cedrene: cedarwood oil is an essential oil derived from various types of conifers such as pine or cypress families. It is a very common ingredient as a cosmetic or food additive ingredient. Various active ingredients are found Cedarwood oil such as sesquiterpene constituents α-cedrene and β-cedrene, cedrol and thujopsene. ALPHABULK utilizes a very high dose of highly purified, isolated α-cedrene which exerts very unique effects compared to all other actives found in cedarwood.

It has recently been identified as a potent agonist of the OR subtype OR10J5, which as previously discussed leads to potent increases in cAMP in target tissues such as skeletal muscle and fat cells. Recent research has shown that α-Cedrene is a promising agent that may be applied to enhance the mass and strength of skeletal muscle.

α-Ionone: Ionones are part of the family of compounds known as rose ketones, they are aroma compounds found in small quantities in a variety of essential oils and fruits such as raspberries. Ionones can be found in three different forms including alpha, beta and gamma. Due to their intense aroma, they are common ingredients in various fragrances, cosmetics and flavoring additives in foods.

ALPHABULK utilizes a massive clinical dosage of a highly purified, isolated α-ionone, which serves a very different and frankly, exciting role. Recent literature has shown that α-ionone is a both potent stimulator of myogenesis and skeletal muscle anti atrophic agent.

How do both α-Cedrene and α-Ionone work?

The mechanism through which they exerts these potent effects is through boosting cAMP, research suggests that they are both potent agonists of OR subtypes OR51E2 and OR10J5, which when expressed, leads to a substantial increase in cAMP levels and subsequent downstream effects.

Both α-Cedrene and α-Ionone has been shown to be highly orally bioavailable and build up in various bodily tissues such skeletal muscle, adipocytes, and the liver, where they can exert its effects where it matters most. They have been shown to affect downstream key anabolic signalling molecules such as PKA Cα, p-CREB, p-FoxO3 and (PGC)-1α1, whilst simultaneously decreasing expression of catabolic signalling molecules such as RING finger 1 (MuRF1) and muscle atrophy F-box (MAFbx), which regulate ubiquitin-proteasome-dependent proteolysis. The amount of p-SMAD2/3 (a downstream molecule of myostatin signalling) significantly diminished, whereas the amounts of IGF-1 signalling molecules including p-AKT, p-mTORC, p-S6K1, and p-4E-BP1 significantly increased in the skeletal muscle of α-ionone-treated subjects. In layman’s terms, key anabolic processes are greatly enhanced while simultaneously key catabolic processes are suppressed, leading to a potent net benefit in promoting anabolism.

Activation of the relevant OR subtypes by both ingredients also exert positive effects in respect to enhancing glucose uptake, increasing insulin sensitivity and research has shown that they lead to reprogramming of metabolic signatures, boosting levels of Adcy3 and of genes involved in fatty acid oxidation, with concomitant downregulation of the ability for fat cells to increase levels of fatty acids! Furthermore cAMP is a vital signalling messenger for the hypothalamic-pituitary-testicular-axis and production of key anabolic hormones such as testosterone, enhanced levels of cAMP directly correlate with enhanced hormone output!

Expect multifaceted benefits from their inclusion ranging from improved increase skeletal muscle anabolism, enhanced anabolic hormone production, increased fat lipolysis and oxidation and increased glycogen synthesis.

Tectochrysin PhytoFUSE: Derived from the roots of Black Ginger, this ingredient is essentially a building block which your body uses as a blueprint to create its own benefit. A study into the various Methoxy Flavones in Black Ginger showed that Tectochrysin was the most potent of the Methoxy Flavones in respect to skeletal muscle hypertrophy, delivering the greatest increases in Myotube diameter. Its beneficial effects include increased protein synthesis and nitrogen retention while promoting other anabolic effects such as increased, muscle endurance, muscle recovery, and lean muscle mass through increasing skeletal muscle hypertrophy.

Additionally, Tectochrysin has been shown to aid in skeletal muscle ATP production, skeletal muscle glucose uptake and exercise performance. PhytoFUSE, is a unique delivery technology that enhances the bioavailability and absorption various classes of compounds, including Methoxy Flavones. Research has shown that this method of delivery can enhance potency and duration of action substantially!

Urolithin B: One of four metabolites of ellagic acid/ellagitannins. Research has shown supplementing the diet with ellagitannins mitigates muscle damage experienced during intense exercise and protects muscle against stresses induced by a high-fat diet. Clinical research on Urolithin B in mice found that it enhanced myotubes growth and differentiation by increasing protein synthesis. It demonstrated an ability to inhibit the ubiquitin–proteasome pathway (UPP), the principal mechanism for protein catabolism. It also induced muscle hypertrophy and reduced muscle atrophy. Signalling analyses indicate a connection between the androgen receptor and the mTORC1 pathway, possibly via AMPK. In studies, UroB at 15µM promoted androgen receptor activity by 90% vs. testosterone at 100µM which promoted androgen receptor activity by 50%. Even at significantly lower doses it outperformed testosterone. Additionally, UroB at 15µM upregulated MPS by 96% while blood glucose at 100 µM upregulated MPS by just 61%. Once again, at a significantly lower dose UroB was more effective in increasing muscle protein synthesis.
